In order to prove the strand separation hypothesis, methods had to be found for the physical separation of parental and daughter DNA molecules. This was first accomplished through use of heavy isotopes such as N15. Bacteria grown in a medium containing the heavy isotope N15 have denser DNA than bacteria grown under normal conditions with N14. Heavy DNA can be separated from light DNA by equilibrium centrifugation in concentrated solutions of heavy salts such as cesium chloride. If DNA replication involves strand separation, definite predictions can be made about the density of the DNA molecules found after various growth intervals in a light medium. After one generation of growth, all the DNA molecules should contain one heavy and one light strand and thus be of intermediate hybrid density. This result is exactly what is observed. EDNAP started life in October 1988 in London when a group of forensic scientists from various European countries came together to find a way of harmonizing the DNA technology for crime investigation. With the thought in mind that an integrated Europe with open borders could well see the escalation of cross-border crimes, there would be a need for forensic scientists to exchange data and compare results of DNA analysis. At the 14th Congress of the International Society for Forensic Haemogenetics (ISFH) in Mainz (1991), EDNAP was accepted as a working group of the ISFH. Within this working group, it is intended that each European country should be represented by one laboratory with sufficient expertise in forensic DNA technology. Leonard Adleman, a mathematician at the University of Southern California, combined microscopic bits of DNA to calculate a well-known computer problem. A DNA computer processes each step comparatively slowly, but it can work on billions of sites at once in a cell or a test tube. The standard computer problem that Adleman tackled in his lab is one that seeks a route connecting seven cities, some of which have no direct connections between them, in such a way that a hypothetical traveller arrives at each city only once without backtracking. He solved this puzzle by treating the chemical units of DNA as computing symbols, then assigning a series of symbols to each "city." He then used DNAs natural affinity to join its strands together to create possible city-to-city sequences. In a test tube, very short bits of DNA zipped on to other bits, until all possible routes to the seven cities were assembled very rapidly. Only amplified ribosomal DNA restriction analysis (ARDRA) was suitable in recognizing different G. vaginalis genotypes. At least 3-4 genotypes were identified with different restriction enzymes, some of which showed a prevalent distribution in certain of the centers from which they were collected. Primed in situ labeling (PRINS) is a fast alternative to traditional in situ hybridization, requiring only a tew handling steps (1,2). In the PRINS reaction, an unlabeled sequence-specific oligonucleotide primer or short DNA fragment is hybridized (annealed) to denatures chromosomal DNA and extended in situ in the presence of hapten-labeled nucleotide triphos phates by a suitable DNA polymerase (3).
