La mutagénèse non dirigée in vitro propose, à partir d’un gène cloné et codant pour une protéine donnée, de construire une collection de formes mutées(de façon aléatoire) de ce gène(librairie allélique). Parmi les variants protéiques codés ainsi obtenus, on peut identifier ceux qui sont complètement inactifs, ceux où la mutation n’ a pas d’effet sur l'activité de la protéine, et enfin ceux pour qui la fonction ou les propriétés physico-chimiques ont été modifiées. Ces derniers peuvent être sélectionnés en fonction d’un critère recherché-thermostabilité par exemple.
