2, record 2, French, - glissement%20antig%C3%A9nique

Heteroduplex is double-stranded DNA in which the two DNA strands do not show perfect base complementarity. When DNA is denatured, the two strands are separated. On renaturation or annealing, complementary DNA strands reassociate and form a homoduplex. However, when there is a mutation in one of the strands then a heteroduplex is formed. The electrophoretic mobility of heteroduplex in polyacrylamide gel is less than that of homoduplex, and they can be detected as an extra slow moving bands. Resolution is better in specially formulated gel matrices (Hydro-link, MDE, etc.). In fragments of under 200 bp insertions, deletions and most single base substitutions are detected. Heteroduplex analysis is mostly used combined with single confirmation analysis (SSCA) on a single gel. 1, record 3, English, - heteroduplex%20analysis

RNA interference (RNAi) is an evolutionarily conserved mechanism in plant and animal cells that directs the degradation of messenger RNAs homologous to short double-stranded RNAs termed "small interfering RNA" (siRNA). The ability of siRNA to direct gene silencing in mammalian cells has raised the possibility that siRNA might be used to investigate gene function in a high throughput fashion or to modulate gene expression in human diseases. 1, record 6, English, - small%20interfering%20RNA

A number of methods are available that can detect known DNA mutations e.g. Allele specific amplification/hybridisation(ASA/H), Oligonucleotide ligation assay (OLA), and mini-sequencing just to mention a few. A common drawback for all of these methods is that they only detect mutations known in advance i.e. mutations outside the small area of detection will go undetected. If the position or nature of mutations is unknown then one must define the type of information required :Screening: Is there a mutation or not? SSCP, DGGE ... How many and where are they localised? Short stretches of RNA have been found attached to the 5' ends of many newly synthesized DNA chains. The starting points for DNA replication may therefore be recognized not by a DNA polymerase, but by a form of RNA polymerase. Following binding of a starter RNA polymerase, a number of ribonucleotides are sequentially linked together until a stop signal is read. The enzyme then detaches leaving a short RNA chain still bound to its DNA template. This RNA chain then serves as a primer onto which DNA polymerase III adds deoxynucleotides. Removal of the RNA primer occurs later through the action of other enzyme(s) which can digest away the RNA component of RNA/DNA hybrids. One enzyme that has this capacity is DNA polymerase I, whose primary task may be to close the gaps between RNA-primed DNA fragments. 1, record 12, English, - RNA%2Dprimed%20DNA%20fragment
