FRAGMENT RESTRICTION [5 records]

Record 1 2012-09-10

English

Subject field(s)
  • Genetics
DEF

Any of the individual polynucleotide sequences produced by digestion of deoxyribonucleic acid with a restriction endonuclease.

French

Domaine(s)
  • Génétique
CONT

English

Subject field(s)

  • Genetics
  • Molecular Biology
DEF

A cloning vehicle which has cleavage sites for two or more restriction endonucleases, thus defining a fragment of DNA which can be removed and replaced by foreign DNA.

CONT

Making more subtle mutations: the "knock in". Although it is often desirable to simply inactivate a gene to determine the importance of a null phenotype, in many cases it is more informative not to inactivate a gene, but rather to modify it so that its function is altered. Again, this is a task (which is the logical equivalent of site directed mutagenesis in a plasmid) that can be solved by homologous recombination. In this case a targeting vector is designed as a replacement vector so that additional genetic sequence are added into the genome. This technique is sometimes called a "knock-in".

DEF

Record 3 2001-11-02

English

Subject field(s)
  • Genetics
DEF

A cloning vehicle which has a single cleavage site for one or more restriction endonucleases, thus allowing the insertion of a fragment of foreign DNA.

CONT

Avipoxviruses, including Fowlpoxvirus (FPV) have been shown previously to infect mammalian cells but are unable to complete their replication cycle. Avipoxvirus based rabies and HIV vaccines have shown promise in preclinical studies. We have begun work to develop the FPV system for application to HPV CTL. We first constructed a new FPV insertion vector (Fig. 3) which facilitates production of recombinant viruses by co-insertion of the gpt marker gene under the control of a vaccinia virus 19K promoter. The gene of interest is inserted into a polylinker downstream of the vaccinia virus 7.5K promoter.

DEF
