In reversible inhibition of enzymes ... the inhibitor participates in a rapidly established, easily reversible equilibrium with the enzyme or enzyme-substrate complex, which can be analyzed in terms of the Michaelis-Menten formalism.... The three major types of reversible enzyme inhibition, competitive, uncompetitive, and noncompetitive, can be experimentally distinguished by the effects of the inhibitor on the reaction kinetics of the enzyme, which may be analyzed in terms of the basic Michaelis-Menten rate equation. For valid kinetic analysis the inhibitor must combine rapidly and reversibly with the enzyme or enzyme-substrate complex. 2, record 2, English, - reversible%20inhibition

3, record 3, French, - inhibition%20non%20comp%C3%A9titive

In competitive inhibition, the binding of inhibitor and substrate to the enzyme is mutually exclusive.... With some proteins, especially regulatory proteins, competitive inhibition can result from the inhibitor binding at another location on the enzyme, thereby altering the configuration at the active site so that substrate cannot bind. Such an enzyme may be thought of as having two different configurations, one that permits binding of substrate. Binding is mutually exclusive and therefore competitive, as in simpler situations where the substrate and inhibitor are competing for binding to the same site. In these more complex examples of competitive inhibition the structure of the inhibitor usually does not resemble that of the substrate. 3, record 5, English, - competitive%20inhibition

According to the symmetry model, binding the first substrate molecule increases the tendency of the remaining subunits to undergo transition to the high-affinity form through an all-or-none effect, all subunits being either in their low-affinity or their high-affinity forms. The sequential model also postulates that the catalytic subunits have two conformational states, but it differs from the symmetry model in postulating that the subunits may undergo individual sequential changes in conformation; between the all-on and all-off states there may be many intermediate conformational states of the enzyme molecule, each having its own intrinsic catalytic activity. 1, record 6, English, - high%2Daffinity%20form

3, record 7, French, - site%20allost%C3%A9rique

Irreversible inhibition occurs when the inhibitor molecule combines irreversibly with the enzyme, chemically modifying its structure and abolishing or greatly reducing its activity. An example is di-isopropylfluorosphosphate (DFP) which inhibits enzymes which have reactive serine groups in their active sites, such as acetylcholinesterase. Thus they act as probes of the active site and as nerve poisons. Another example is heavy metals such as lead or mercury which react with SH groups in the active site of enzymes. 1, record 9, English, - irreversible%20inhibition
