Les composés élués de la colonne sont dirigés dans un plasma hélium ou argon excité par microonde. Le spectre atomique produit par le soluté est analysé par un monochromateur réglé sur la longueur d’onde de l'élément recherché. Le signal délivré par le photomultiplicateur situé à la sortie du monochromateur est amplifié puis enregistré. À la différence d’un photomètre de flamme, de détecteur met en œuvre des énergies plus importantes qui fragmentent les molécules. Les fragments sont excités, leur spectre d’émission est caractéristique de l'élément.
