La première étape consiste à fabriquer un anticorps monoclonal capable de reconnaître la molécule étudiée(dans ce cas, le récepteur l'IL2). A cette fin, ces chercheurs immunisèrent des souris avec des lymphocytes T humains activés par un antigène, et recherchèrent les anticorps ne réagissant qu'avec les lymphocytes T activés et non avec les lymphocytes au repos. Ces anticorps furent baptisés "anti-TAC"(Anti T Activated Cells).
