Il est risqué de semer de la luzerne là où cette plante a déjà poussé parce que les vieux peuplements produisent une toxine qui ralentit la germination et la croissance des nouvelles pousses. On appelle ce phénomène l'autotoxicité. L'établissement de la culture peut être difficile, à moins que le peuplement existant ait subi un labourage ou une pulvérisation au moins trois semaines avant les nouveaux semis. Ces toxines peuvent persister jusqu'à six mois, ce qui suffit à compromettre de façon permanente le rendement des nouveaux peuplements. Pour assurer un rendement maximal, il faut cultiver une autre culture pendant un an. Les toxines ne sont pas présentes la première année dans les nouveaux peuplements.
