De nouvelles machines à PCR ont été conçues de façon à permettre la détection des produits d’amplification extemporanément à leur synthèse. Les méthodes de détection qu'ils mettent en œuvre sont basées sur la liaison à l'ADN, durant la réaction, de marqueurs fluorescents ou de l'activité 5’nucléase des ADN polymérases thermostables. Ce dernier type de PCR peut être qualifié de PCR par déplacement de coupure(nick translation PCR). Polymers, because of their structure, additives or structure defects, can contain what are referred to as chromophor groups, which by absorbing UV-light can increase the electronic energy level of the polymer. The polymer in this situation can readily react further, for example, with oxygen (photo-oxidation). The higher scale energy of the polymer can be transferred to the quencher by adding energy quencher.
