The polymerase chain reaction (PCR) is a method of copying a DNA or RNA strand thousands of times, in a matter of minutes, by using the natural ability of an enzyme called polymerase to copy DNA. ... Scientists must know ... what DNA sequences flank or frame either ends of the desired portion. Complementary copies of the DNA sequences that flank the ends are created by using special enzymes; they are called primers. They identify the start and end of the copying process. ... Pronounced variation in the number and location of edited sites was found among species. Most species had a relatively large number of edited sites (from 25 to 34). However, no RNA editing of the coxI sequence was found in Gingko biloba or Larix sibirica. The sequence composition of the investigated coxI fragment suggests that the coxI gene in G. biloba and L. sibirica originated from edited mitochondrial coxI transcripts by reverse transcription followed by insertion into the nuclear genome or back into the mitochondrial genome. Our results also demonstrate that where there a large number of edited sites, RNA editing can accelerate the divergence of sequences among species. 2, record 9, English, - RNA%20editing

1, record 10, French, - mutag%C3%A9n%C3%A8se%20dirig%C3%A9e%20%C3%A0%20l%26rsquo%3Baide%20d%26rsquo%3Bun%20oligonucl%C3%A9otide%20synth%C3%A9tique

Since alleles derived from different parental origins that are identical in nucleotide sequence may exhibit imprinting, the differences between the paternal and maternal genetic material are due to differential programming during spermatogenesis or oogenesis. The different packaging constraints of the sperm cell compared with the ovum also may be important. The regulatory mechanisms superimpose on the nucleotide sequence of a gene to determine the location, timing and degree of expression, and are termed epigenetic effects. Epigenetic modification (rather than genetic divergence of the gametes) of the DNA is likely to be the signal that marks the sperm and egg alleles as different. Imprinting is a special type of epigenetic inheritance that is passed through the gene line into somatic tissues of the progeny and is parent-specific. 1, record 11, English, - epigenetic%20modification

Interferons are a family of naturally occurring small proteins that are produced and secreted by cells in response to viral infection. Alpha and beta interferons are classified type 1 interferons and include more than 25 different molecules. Consensus interferon (CIFN) is a novel, recombinant type 1 interferon containing 166 amino acids. CIFN was derived by scanning the sequences of natural alpha interferons and assigning the most frequently observed amino acid in each corresponding position. CIFN, when compared on an equal mass basis with interferon (IFN) alfa-2 and alpha-2b in in vitro assays, typically displays 5-10 tiems higher biological activity. 1, record 14, English, - Consensus%20Interferon

Primed in situ labeling (PRINS) is a fast alternative to traditional in situ hybridization, requiring only a tew handling steps (1,2). In the PRINS reaction, an unlabeled sequence-specific oligonucleotide primer or short DNA fragment is hybridized (annealed) to denatures chromosomal DNA and extended in situ in the presence of hapten-labeled nucleotide triphos phates by a suitable DNA polymerase (3). The technique is especially suited for the rapid identification of chromo somes with oligonucleotide primers specific for individual chromosomal sequences and chromosomal elements like centromer or satellite sequences. 2, record 16, English, - primed%20in%20situ%20labeling%20of%20nucleic%20acids

Multiplex DNA sequencing: The increasing demand for DNA sequences can be met by replacement of each DNA sample in a device with a mixture of N samples so that the normal throughput is increased by a factor of N. Such a method is described. In order to separate the sequence information at the end of the processing, the DNA molecules of interest are ligated to a set of oligonucleotide "tags" at the beginning. The tagged DNA molecules are pooled, amplified, and chemically fragmented in 96-well plates. The resulting reaction products are fractionated by size on sequencing gels and transferred to nylon membranes. These membranes are then probed as many times as there are types of tags in the original pools, producing, in each cycle of probing, autoradiographs similar to those from standard DNA sequencing methods. 1, record 19, English, - multiplex%20DNA%20sequencing
