On peut sélectionner à partir de banques combinatoires, des oligonucléotides présentant une spécificité et une affinité élevées pour un grand nombre de molécules. Cependant, leur sensibilité aux nucléases limite leur utilisation. La synthèse chimique de L-oligonucléotides échappant pour cause de sélectivité stérique à l’hydrolyse enzymatique, permet de contourner ce problème. La conception de tels ligands, dénommés «spiegelmers» par l’équipe allemande de Jens Fürste, où est basée sur la sélection in vitro (SELEX) de D-ARN présentant une affinité pour les inverses optiques des cibles visées.
