The dominant lethal test is based on the preimplantation loss of eggs or on the formation of dead embryonic implants following the injection of mutagens into a male or female mouse at a specific time before mating (Bateman and Epstein, 1971). Le test de létalité dominante a tout d’abord été utilisé avec succès pour mesurer les effets mutagènes des radiations ionisantes par W. Russel et al. [...] puis par Bateman. Il a depuis lors été étendu à d’autres mammifères tels que le rat et le hamster. Dans sa forme initiale, il consiste tout d’abord à croiser les animaux préalablement traités(l'un ou l'autre sexe ou les deux) en repérant exactement le moment de la fécondation par une technique adéquate qui dépend de l'espèce animale en cause. Les femelles sont ensuite sacrifiées au cours de la seconde moitié de la période de grossesse à un moment judicieusement choisi et toujours le même. On relève ensuite le nombre d’embryons en vie et le nombre de corpora lutea de grossesse qui correspond en règle générale, au nombre d’ovules effectivement fécondés. On note ensuite le nombre de déciduomes qui correspond à la mortalité après implantation intra-utérine des jeunes embryons.
