L'unité d'activité enzymatique la plus utilisée est définie comme la quantité d'enzyme qui produit la transformation d'1,0 micromole (10[exposant-6] mole) de substrat par minute à 25° C, dans les conditions optimales de dosage. L'activité spécifique est le nombre d'unités d'activité enzymatique par mg de protéine. C'est une mesure du degré de pureté de l'enzyme, car elle augmente au cours de la purification et atteint une valeur maximum et constante lorsque l'enzyme est pur.
