Myosin molecules were known to be more complex: they have globular "head" domains that bind actin filaments and helical "tail" domains that self-associate to form bipolar myosin filaments. In 1963 Hugh E. Huxley of the MRC demonstrated that myosin heads bind to actin filaments at an acute angle; under the electron microscope, the filaments, adorned with myosin heads, looked like a series of arrowheads.

Les molécules de myosine ont une tête globulaire, qui se lie aux filaments d'actine, et une queue hélicoïdale. Les queues s'associent spontanément entre elles pour former des filaments de myosine bipolaires [...] En 1963, Hugh Huxley, du Conseil britannique de la recherche médicale, montra que les têtes des molécules de myosine se lient aux filaments d'actine en se couchant légèrement contre eux; au microscope électronique à balayage, on voit que les filaments d'actine auxquels sont liées les têtes de myosine forment des flèches emboîtées, de sorte que les filaments d'actine sont naturellement orientés, avec une partie «barbée» et une partie «pointue».
