En chimie, l'encapsulation consiste à enfermer une substance dans une capsule. L'objectif est : - soit de protéger la substance contre des interactions avec le milieu extérieur. On encapsule par exemple certains filtres [ultraviolets] UV contenus dans les crèmes solaires pour les stabiliser et les empêcher d'irriter la peau; - soit de transporter la substance jusqu'à l'endroit où elle sera libérée pour agir. Les liposomes par exemple sont des sphères utilisées pour transporter les substances actives des cosmétiques dans la peau. Selon la taille des capsules, on parle de microencapsulation (dimension de l'ordre du micromètre) ou de nanoencapsulation (dimension de l'ordre du nanomètre).

An immobilization method by which cells or enzymes are enveloped inside semi-permeable membranes which allow the passage of substrate and product molecules, but which do not allow the enzyme to pass.

Various methods for immobilizing enzyme molecules have been examined. Physical adsorption and chemical bonding of enzyme molecules to solid surfaces have been studied extensively, but these techniques often cause inactivation of the enzyme and, in extreme cases, denaturation. Encapsulation without chemical reactions would seem to be the best way to avoid these problems.

[A microencapsulation process] similar to the liquid surfactant membrane has been developed which gives a permanent membrane [Kitajema et al. 1969). This, so-called, liquid drying method is carried out by emulsifying an aqueous solution of the enzyme in an organic solvent which has a boiling point lower than that of water (usually benzene, cyclohexane, or chloroform). The organic solvent also contains the membrane-forming polymer and surfactants. The emulsion is then dispersed in an aqueous medium, which contains protective colloidal substances (gelatin, polyvinylchloride) and surfactants, to give a second emulsion. Removal of the organic solvent by, for example, rotary evaporation from the second emulsion results in the formation of the microencapsulated enzymes.

OBS

The major advantage of this procedure is that no deactivation of the enzyme can occur as a result of monomer interactions due to the preformed polymer being used. Disadvantages of the procedure include the low yields of the microcapsules produced, due to problems of preparing the second emulsion, and the time required to completely remove the organic solvent which is essential for membrane production.
