Atomic-force microscopy is similar to scanning-tunneling microscopy (STM) in that it can image surfaces at atomic-scale resolution. The difference between AFM and STM is that AFM does not require that the sample be an electrically conducting material. Like STM, it uses an atomically-sharp tip that is brought very close to the surface. The tip will feel a chemical attraction or repulsion and will move up or down on its supporting cantilever. This movement is monitored by an optical laser beam as the tip is moved laterally across the surface. La microscopie à force atomique (AFM : «atomic force microscopy») a été introduite en 1986 par G. Binnig, C.F. Quate et C. Gerber, comme une application du concept de microscope à effet tunnel (STM : «scanning tunneling microscope») [...] permettant l'étude de surfaces de matériaux isolants à l'échelle atomique. En combinant les principes du microscope à effet tunnel et du stylet profilométrique, les auteurs démontraient la possibilité d'imager, à l'air libre, la surface d'échantillons conducteurs ou non, avec une résolution latérale de 30 Å [angström] et une résolution verticale inférieure à 1 Å. La technique a, depuis lors, été adaptée à différents environnements tels que le vide, le milieu liquide, les basses températures, les champs magnétiques et aussi pour des applications en chimie ou en biologie. L'AFM est basée sur la mesure des forces entre un fin stylet et la surface étudiée. Le capteur de force est un ressort-lame (stylet) encastré à une extrémité et muni d'une pointe à l'autre extrémité; il est encore appelé «cantilever». Les forces d'interaction modifient la déflection ou la torsion statique ou oscillante du stylet. La mesure des déformations du «cantilever» dans les microscopes de force actuels s'effectue, le plus souvent, grâce à la déviation d'un faisceau lumineux («diode laser») réfléchi par l'extrémité du stylet, méthode proposée dès 1988 par G. Meyer et N. La [...] microscopie à effet tunnel (STM) a été inventée par G. Binning et H. Rohrer en 1981. Son principe est le même que la microscopie optique à champ proche. Une sonde métallique est approchée au plus près de la surface d'un matériau conducteur, afin de détecter les ondes électroniques évanescentes des électrons de conduction du métal. On sait que l'amplitude de ces ondes décroît exponentiellement en fonction de la distance à la surface pour un observateur placé en dehors du métal. Le courant tunnel sonde-surface est donc une fonction décroissante de la distance qui sépare la surface de l'apex de la pointe : typiquement un ordre de grandeur par 0,1 nm [nanomètre]. De plus, le courant tunnel reste très localisé au voisinage de la pointe. Near infrared microscopy has shown potential to discriminate bone particles of fish origin and those of terrestrial animal origin in the sediment fraction of feedstuffs. In this study, the use of this method for the detection of processed animal proteins in the flotate is investigated. La microscopie en proche infrarouge (NIRM) a démontré son potentiel pour discriminer les particules d'os de poisson de celles des animaux terrestres dans la fraction sédimentée des aliments composés pour animaux. L'identification de traces de protéines animales transformées (PAT) par cette technique est basée sur l'évaluation des spectres obtenus à partir des particules individuelles présentes dans l'échantillon. deconvolution microscope : using the deconvolution microscope, researchers can see the locations of target molecules inside the cell. Seeing where glowing molecules reside within a cell is helpful for a range of research projects -- from studies of how the distribution of various proteins changes during an organism's development to investigations of how administration of a drug alters the locations of certain proteins. The microscope, called DeltaVision, is manufactured by Applied Precision, based in Issaquah, Wash. It works by magnifying images and quantifying the intensity of light shed by the sample. The user specifies which colors of light are of interest. For instance, a user can program the microscope to report the intensity of a particular shade of green shining from the cytoplasm of a cell. After a researcher uses the microscope to make a series of optical slices through a specimen, the scope's computer "deconvolves" the images -- in other words, it subtracts the out-of-focus light.Then the computer stacks together the images of the slices to reconstruct the original microscope sample. Confocal microscopy. Unlike "normal" microscopy, only a spot of a minimum size in the specimen is illuminated in confocal microscopy. To enable an image to be formed from this, the specimen must be scanned. The image of the spot is directed through a pinhole stop in an intermediate image plane. As a result, only light from the focal plane can reach the detector (a photomultiplier). All other (out-of-focus) planes are blocked out. This results in an "optical section". The images are stored electronically and displayed on a monitor. A series of optical sections can be recorded by moving a motor a slight distance along the z-axis each time an image has been recorded, after which the next image is then also recorded. Such a z-series permits the electronic reconstruction of the three-dimensional structure using suitable computer programs. Cette technique, initialement introduite par Castaing et Slodzian dès 1962 pour l'analyse de surface en géologie et en métallurgie, a été appliquée à la biologie par Galle à partir de 1970. Le principe de base de la méthode repose sur un principe simple; si une surface est bombardée par un faisceau d'ions primaires énergétiques ces particules vont induire la rupture des liaisons chimiques des molécules présentes dans les couches superficielles de l'échantillon et conduire à la désorption d'atomes ou de petits fragments polyatomiques neutres ou chargés (ions secondaires). Ces ions peuvent alors être accélérés et focalisés par des lentilles électrostatiques pour finalement être analysés et «triés» par un spectromètre de masse. Scanning transmission electron microscopy (STEM) combines the principles of transmission electron microscopy and scanning electron microscopy and can be performed on either type of instrument. Like TEM [transmission electron microscopy], STEM requires very thin samples and looks primarily at beam electrons transmitted by the sample. One of its principal advantages over TEM is in enabling the use of other signals that cannot be spatially correlated in TEM, including secondary electrons, scattered beam electrons, characteristic X-rays, and electron energy loss. Like SEM [scanning electron microscopy], the STEM technique scans a very finely focused beam of electrons across the sample in a raster pattern. Interactions between the beam electrons and sample atoms generate a serial signal stream, which is correlated with beam position to build a virtual image in which the signal level at any location in the sample is represented by the gray level at the corresponding location in the image. Laser Scanning Confocal Microscopy (LSCM, also referred to as CSLM, Confocal Scanning Laser Microscopy) is now established as a valuable tool for obtaining high resolution images and 3-D reconstructions of a variety of biological specimens. In LSCM, a laser light beam is expanded to make optimal use of the optics in the objective. Through a x-y deflection mechanism this beam is turned into a scanning beam, focussed to a small spot by an objective lens onto a fluorescent specimen. The mixture of reflected light and emitted fluorescent light is captured by the same objective and (after conversion into a static beam by the x-y scanner device) is focused onto a photodetector (photomultiplier) via a dichroic mirror (beam splitter). The reflected light is deviated by the dichroic mirror while the emitted fluorescent light passes through in the direction of the photomultiplier. A confocal aperture (pinhole) is placed in front of the photodetector, such that the fluorescent light (not the reflected light!) from points on the specimen that are not within the focal plane (the so called out-of-focus light) where the laser beam was focussed will be largly obstructed by the pinhole. Infrared microscopy is a specialized branch of optical (light) microscopy. One of the most important new designs in the field of laser scanning microscopy is Bio-Rad's new multiphoton microscope. The primary benefit of multiphoton or two-photon microscopy is that it can penetrate much deeper into tissue samples, permitting an unobstructed view of cells such as neurons. The way it accomplishes this feat involves an unusual characteristic of light. When two photons hit a fluorophor simultaneously, it's as if one photon of half the wavelength excited the targeted fluorophor. If two photons of infrared light are directed at a particular sample, they will penetrate to a depth of several millimeters and then combine in the focal plane where the photons are most concentrated, reacting as if one photon of blue light hit the sample. The result is a deep tissue image of superior resolution.
