Scanning tunnelling microscopy (STM) has proved to be a ... powerful technique in the field of surface science. The fact that sets the STM apart from most other surface sensitive techniques is its ability to resolve the structure of surfaces on an atomic scale, that is atom-by-atom, and furthermore its ability to study the dynamics of surface processes. 4, record 1, English, - scanning%20tunneling%20microscopy

Record number: 1, Textual support number: 1 CONT

In scanning electron microscopy (SEM), a fine probe of electrons with energies typically up to 40 keV [kiloelectron volts] is focused on a specimen, and scanned along a pattern of parallel lines. Various signals are generated as a result of the impact of the incident electrons, which are collected to form an image or to analyse the sample surface. These are mainly secondary electrons, with energies of a few tens of eV [electron volts], high-energy electrons backscattered from the primary beam and characteristic X-rays. 5, record 4, English, - scanning%20electron%20microscopy

Scanning transmission electron microscopy (STEM) combines the principles of transmission electron microscopy and scanning electron microscopy and can be performed on either type of instrument. Like TEM [transmission electron microscopy], STEM requires very thin samples and looks primarily at beam electrons transmitted by the sample. One of its principal advantages over TEM is in enabling the use of other signals that cannot be spatially correlated in TEM, including secondary electrons, scattered beam electrons, characteristic X-rays, and electron energy loss. Like SEM [scanning electron microscopy], the STEM technique scans a very finely focused beam of electrons across the sample in a raster pattern. Interactions between the beam electrons and sample atoms generate a serial signal stream, which is correlated with beam position to build a virtual image in which the signal level at any location in the sample is represented by the gray level at the corresponding location in the image. Its primary advantage over conventional SEM imaging is the improvement in spatial resolution. 3, record 5, English, - scanning%20transmission%20electron%20microscopy

SNOM is the acronym for "scanning near-field optical microscopy," an alternative name for NSOM (near-field scanning optical microscopy). The basic principle of such a kind of technique is quite simple: light passes through a sub-wavelength diameter aperture and illuminates a sample that is placed within its near field, at a distance much less than the wavelength of the light. The resolution achieved is far better than that which conventional optical microscopes can attain. 4, record 6, English, - scanning%20near%2Dfield%20optical%20microscopy

Microscopists are now in a position to transfer the refinements of the past 100-year techniques for enhancing contrast and highlighting specific features, such as polarization contrast, immunofluorescence and phase contrast to scanning near-field optical microscopy. With existing scanned-probe microscopes we can "see" the nanometer realm of molecules and microcircuits. As the same technology is applied to optical microscopy, we may eventually come to see that world in the familiar terms of light, shadow and color. 6, record 6, English, - scanning%20near%2Dfield%20optical%20microscopy

Laser Scanning Confocal Microscopy (LSCM, also referred to as CSLM, Confocal Scanning Laser Microscopy) is now established as a valuable tool for obtaining high resolution images and 3-D reconstructions of a variety of biological specimens. In LSCM, a laser light beam is expanded to make optimal use of the optics in the objective. Through a x-y deflection mechanism this beam is turned into a scanning beam, focussed to a small spot by an objective lens onto a fluorescent specimen. The mixture of reflected light and emitted fluorescent light is captured by the same objective and (after conversion into a static beam by the x-y scanner device) is focused onto a photodetector (photomultiplier) via a dichroic mirror (beam splitter). The reflected light is deviated by the dichroic mirror while the emitted fluorescent light passes through in the direction of the photomultiplier. A confocal aperture (pinhole) is placed in front of the photodetector, such that the fluorescent light (not the reflected light!) from points on the specimen that are not within the focal plane (the so called out-of-focus light) where the laser beam was focussed will be largly obstructed by the pinhole. In this way, out-of-focus information (both above and below the focal plane) is greatly reduced. 2, record 9, English, - confocal%20laser%20scanning%20microscopy

Confocal laser scanning microscopy with 1-photon excitation ... became a practical technique with the introduction of the first commercially available models in the early 1980's. 2, record 10, French, - microscopie%20confocale%20%C3%A0%20balayage%20laser%20%C3%A0%20excitation%20monophotonique
