On étudie trois groupes de sujets recevant le placebo ou la substance de référence et la nouvelle substance. A l'issue de l'étude, on compare les résultats obtenus dans ces trois groupes ... cet essai se déroule " à l'aveugle " : on distingue le simple aveugle où le malade ignore le médicament qu'il a reçu, mais le médecin connaît le médicament et le double aveugle où l'effet est jugé par le malade ou le médecin, mais ni l'un ni l'autre ne sait ce qu'il a reçu ou donné.
