Le B17 avait été conçu comme un bombardier de jour. Il devait donc disposer d'un armement défensif beaucoup plus important que celui d'un bombardier de nuit. L'appareil était véritablement hérissé de mitrailleuses de fort calibre (12,5 mm) qui justifiaient son surnom de «forteresse volante».
