Because of specific base pairing, if the sequence of one chain (for example, TCGCAT) is known, that of its partner (AGCGTA) is also known. The opposing sequences are referred to as complementary, and the corresponding polynucleotide partners as complementary chains. La molécule ADN est composée de deux chaînes polynucléotidiques complémentaires liées par des liaisons hydrogènes, ce qui implique l'existence de ponts hydrogènes spécifiques. Lors de la réplication chaque chaîne a un rôle matriciel, ce qui implique un mode de réplication semi-conservatif.
